How can uMelt help? Users of uMeltSM know the software provides accurate high-resolution melting curve predictions. Users also know that running a PCR product on agarose gel has been the gold standard to check PCR products. What some individuals may not know is that uMelt can also provide validation for experiments when an apparent contradiction exists between gel and melting data. In the example below, all three figures are correct. The gel indicates that only PCR products of one size are present. However, due to differing GC-content distribution along the sequence, multiple melting transitions are present. This is not a contradiction. Single PCR products (indicated by the gel) can have multiple melting domains (indicated by the experimental and predicted melting curves).
